Effect Of The Structure On The Binding Of Nickel Schiff Base Complexes To Dna

Three Schiff base complexes with different Structure were selected as: 1- N, N’ Bis [3- tert-butyl-5-[triphenyl- phosphonium – methyl) - salicylidene] 1,2 ethylene- diamine nickel(III) acetate. 2- N, N’ Bis [3- tert-butyl-5-[triethyl- amonium – methyl) - salicylidene] 1,2 ethylene- diamine nickel(II) acetate. 3- N, N’ Bis [3- tert-butyl-5-[triethyl- amonium – 1,8 anthraquine) - salicylidene] 1,2 ethylene- diamine nickel(II) acetate. The complexes were synthesized. Then, the interaction of complexes with DNA were investigated by UV-vis Linear dichroism spectroscopy. The absorption spectra of complexes at various concentrations of DNA showed hypochromism and red shift. Moreover the results exhibited that the structure of complexes specifically on the bridge was more important for interaction with DNA. While in the Salen Ni complexes; the presence of the bulky groups on both side of complex increases the affinity of the complex for DNA. But presence of less bulky group is more effective on the LD experiment. Finally, the binding constant of the complexes were compared together and the results compared to complement the affinity of binding. Keyword: DNA binding, Schiff base, Linear Dichroism
