Performance Of Cellulase Enzymes From Trichoderma Reesei And Aspergillus Niger To Decompose Corncob Into Glucose: Effects Of Incubation Time And Ratios Of Additional Microbes

The research on the performance of cellulose enzymes from Trichoderma reesei and Aspergillus niger to decompose corncobs into glucose with the influence of incubation time and the ratio of the addition of microbes has been done. This study aims to learn the effect of incubation time and the ratio of the addition of microbes of Trichoderma reesei and Aspergillus niger during hydrolysis of corn cobs into glucose. This study uses Solid State Fermentation. This study begins with the physical pretreatment of raw materials by downsizing up to 50 mesh. Pretreatment is aimed to increase the surface area of contact and break down of lignin which envelops cellulose. Trichoderma reesei and Aspergillus niger were cultured in Potato Dextrose Agar medium for 72 hours at the temperature of 20 oC. Then, it is inoculated in Mandel Weber's Medium for 48 hours at the temperature of 20 oC. Variation of parameters are the effect of incubation time (3, 4, 5, and 7 days) and the ratio of the addition of microbes (1:0, 0:1, 1:1, 1:2 and 2:1) (v/v). Trichoderma reesei : Aspergillus niger (2:1) with 5 days of incubation gave the best result than others.
